Effect: It unlocks the level 'Island Infiltration'.

Pause the game during Enemies Vanquished level.
How:
Hold Left Trigger and press Right, Right, Left, Left, Up.

How:
Rather than jumping into the cable cars and leaving yourself vulnerable to damage, just jump on top of the cable car cables which, unlike real cable car cables, are actually completely solid, like a ledge. You can then snipe or shoot from above easily, or even run from one cabin to the other with the flag in this way.
